How Retatrutide Works
Retatrutide (LY3437943) is an investigational polypeptide that simultaneously engages three metabolically relevant receptors: GLP-1R, GIPR and the glucagon receptor.
GLP-1R activation enhances glucose-dependent insulin secretion and slows gastric emptying. GIPR co-agonism amplifies insulin release while modulating adipose signalling. Glucagon receptor engagement — usually associated with hyperglycemia — is balanced by the incretin activity and is investigated for its lipolytic and energy-expenditure effects.
In preclinical and early clinical research, this triple agonism has produced striking metabolic outcomes. Studies remain ongoing.
